The European sea bass, Dicentrarchus labrax L., tolerates a range of salinities from freshwater to hyper-saline. To study differences in protein expression, fish were reared in both freshwater and seawater. After 3-month acclimation, gill and intestine epithelia were collected and the soluble protein extracted. In all, 362 spots were differentially expressed in the gills and intestines of fishes reared in seawater compared to those from freshwater. Fifty differential protein spots were excised from a colloidal Coomassie-stained gel. Nine separate protein spots were identified unambiguously by mass spectrometry and database searching. Among the six proteins over-expressed in gill cells in seawater, five were cytoskeleton proteins and one was the aromatase... |

Objectives : Presently, most of marine fish farmers around the Mediterranean basin grow genetically wild fish strains, which have not been genetically improved for culture performance (growth, appearance, disease resistance etc.). It is inevitable that the present gap for genetically improved strains of marine fish, and their reliable supply, will be filled. It will ultimately dominate the competitive component of marine culture industries once they evolve into commodity-style agribusiness (with fish prices being set at the lowest possible cost of production and the best quality). Those countries, which can competitively develop and supply the improved strains, will enjoy the benefit (perhaps survival) for their local industries. They will avoid... |

This paper presents a non invasive, rapid and reliable way to quantitatively assess fin erosion in sea bass (Dicentrarchus labrax). The method is based on a visual assessment of fin profile and area loss of all fins except the anterior dorsal, on a scale from 0 to 4 in comparison with a perfect fin. The effects of stocking density (SD) and temperature on fin damage were investigated under experimental conditions (100-250 g fish). Over a 4-month period, mean erosion index (mean erosion level of all fins) was 10 times higher at 120 than at 20 kg m(-3), where most fins were undamaged. Damage was also dependent on time and oxygen concentration (35% lower at 53% O-2 saturation than at 105%). Fin condition was also affected by temperature: mean erosion index was... |

The optimal conditions for the retention of the second polar body in sea bass eggs were investigated by altering the timing, intensity and duration of application of pressure and cold shocks. Treatment optima for cold shocks were 0-1 degrees C for 15-20 min at 5 min after fertilisation (a.f.) and 8500 psi for 2 min at 6 min a.f. for pressure shocks. Meiogenesis was obtained by fertilising eggs with UV-irradiated homologous sperm (32,000 erg mm(-2)) and pressure or cold shocking eggs as above. 100% triploidy was induced following definition of liable periods for the disruption of the meiotic spindle obtained in gynogenesis. Ploidy investigations were performed on experimental groups by flow-cytometry. Verification of uniparental transmission in meiogens was... |

Although the actiology of swimbladder inflation anomalies in important aquaculture species such as European sea bass D. labrax is not fully determined, culture conditions are commonly suggested as main contributory factors. Little information is available on whether swimbladder inflation has a genetic basis for its expression too. In this work, 24 full-sibling sea bass families from a 4 dams x 6 sires factorial crossing were reared under communal conditions. The larvae developing normal and abnormal (uninflated or hyper-inflated) swimbladders were genotyped at four microsatellite loci, Labrax-3, Labrax-13, Labrax-17, Labrax29, and allocated to the individual breeders. Out of 273 offspring, 97% could be assigned to a single parental pair. The genotype and... |

Melatonin contributes to synchronizing behaviors and physiological functions to daily and seasonal rhythm in fish. However, no coherent vision emerges because the effects vary with the species, sex, moment of the year or sexual cycle. And, scarce information is available concerning the melatonin age, receptors, which is crucial to our understanding of the role melatonin plays. We report here the full length cloning of three different melatonin receptor subtypes in the sea bass Dicentrarchus labrax, belonging, respectively, to the MT1, MT2 and Mellc subtypes. MT1, the most abundantly expressed, was detected in the central nervous system, retina, and gills. MT2 was detected in the pituitary gland, blood cells and, to a lesser extend, in the optic tectum,... |

When juvenile turbot Scophthulmus maximus and sea bass Dicentrarchus labrax were fed to satiation, growth and food intake were depressed under hypoxia (3.2 +/- 0.3 and 4.5 +/- 0.2 mg O-2 l(-1)). However, no significant difference in growth was observed between fishes maintained in hypoxia and fed to satiation and fishes reared in normoxia (7.4 +/- 0.3 mg O-2 l(-1)) and fed restricted rations (same food intake of fishes at 3.2 mg O-2 l(-1)). Routine oxygen consumption of fishes fed to satiation was higher in normoxia than in hypoxia due to the decrease in food intake in the latter. Of the physiological parameters measured, no significant changes were observed in the two species maintained in hypoxia. This study confirms the significant interaction between... |

The present study employed a patch clamp technique in isolated seabass ventricular myocytes to investigate the hypothesis that oleic acid (OA), a mono-unsaturated fatty acid, can exert direct effects upon whole-cell barium currents. Acute application of free OA caused a dose-dependent depression of the whole-cell barium current that was evoked by a voltage step to 0 mV from a holding potential of -80 mV. The derived 50% inhibitory concentration (IC50) was 12.49 +/- 0.27 mu mol l(-1). At a concentration of 30 mu mol l(-1), OA significantly reduced the current density to about 45% of control values, but did not modify either the shape of the current-density voltage relationship or the apparent reversal potential. In addition, OA did not modify the voltage... |

In two experiments, a computerized on-demand feeding system coupled with a PIT tag monitoring device was used to continuously record the triggering activity by ca. 50 individual sea bass for 55 days (Exp. 1, initial average body weight and coefficient of variation, CV: 299 g, 15%) and 69 days (Exp. 2, 157 g, 13%). Each group was stocked in 1 m(3) tanks and exposed to a water temperature of 21 +/- 1 degrees C and a light regime of 16:8 LD. Only 5% of the triggering activity was not associated with simultaneous PIT tag detection. Although each individual was detected via PIT tag during the experiment, only 67% and 74% of the fish actuated the trigger at least once in Exp. 1 and Exp. 2, respectively. Moreover, only two fish in Exp. 1 and one fish in Exp. 2... |

Euryhaline teleosts possess the capacity to osmoregulate under various environmental conditions (freshwater to hypersaline water). This physiological capacity is generally monitored using enzyme activity assays (Na+/K+-ATPase...), hormones quantification (prolactine, growth hormone) or their mRNAs expression. To date, few studies addressed the genetic correlates of adaptation to varying salinity at a molecular level in such fish. In the sea bass Dicentrarchus labrax, genetic differentiation was observed at specific allozyme loci between lagoon- and open-sea populations. In the present study, we investigated transcriptomic response of D. labrax to salt- and freshwater acclimation in two organs involved in osmoregulation, gill and intestine. By using... |

Climate change challenges the capacity of fishes to thrive in their habitat. However, through phenotypic diversity, they demonstrate remarkable resilience to deteriorating conditions. In fish populations, inter-individual variation in a number of fitness-determining physiological traits, including cardiac performance, is classically observed. Information about the cellular bases of inter-individual variability in cardiac performance is scarce including the possible contribution of excitation-contraction (EC) coupling. This study aimed at providing insight into EC coupling-related Ca2+ response and thermal plasticity in the European sea bass (Dicentrarchus labrax). A cell population approach was used to lay the methodological basis for identifying the... |

Cardiac preload (central venous; pressure, Pcv), mean circulatory filling pressure (MCFP), dorsal aortic blood pressure (P-DA) and relative cardiac output (Q) were measured in sea bass (Dicentrarchus labrax) at rest and while swimming at 1 and 2 BL s(-1). MCFP, an index of venous capacitance and the upstream venous pressure driving the return of venous blood to the heart, was measured as the plateau in Pcv during ventral aortic occlusion. Compared with resting values, swimming at 1 and 2BL s(-1) increased Q (by 15 +/- 1.5 and 38 +/- 6.5%, respectively), Pcv (from 0.11 +/- 0.01 kPa to 0.12 +/- 0.01 and 0.16 +/- 0.02 kPa, respectively), MCFP (from 0.27 +/- 0.02 kPa to 0.31 +/- 0.02 and 0.40 +/- 0.04 kPa, respectively) and the calculated pressure gradient for... |

The influence of dietary TAG source (fish oil, triolein, and coconut oil) and level (7.5 and 15% of the diet) on growth, lipase activity, and mRNA level was studied in sea bass larvae, from mouth opening until day 24 and from day 37 to 52. Fish oil and triolein induced better growth in both experiments, this being significant at a higher dietary level. Coconut oil significantly decreased growth at the higher level, possibly as the result of an excessive supply of medium-chain TAG. Growth was not related to lipase specific activity, suggesting a production in excess to dietary needs. Body lipid content was positively related to dietary lipid level and was affected by lipid quality. In addition, larval FA composition generally reflected that of the diet. The... |

A sperm cryopreservation protocol adapted from turbot, was tested on sea bass using either 250-mu L straws or 1.5-mL cryovials. A dilution to 1/3 in Mounib's extender and a cooling rate of 65 degrees C.min(-1) allowed frozen sperm to recover an initial motility similar to that of fresh sperm at thawing; however, significant differences in motility (P < 0.001, n = 10 fish semen) were observed at further post-activation times, the motility decrease being faster in thawed sperm. At the experimental scale, triplicate inseminations of 2-mL aliquots (approximately 2 000 eggs) showed a significant fertility decay of thawed sperm compared to that of fresh sperm (P < 0.01, n = 12 fish semen) when a discriminating 35.10(3) spermatozoa to egg ratio was applied.... |

During 84 days, the effects of density on juvenile sea bass (Dicentrarchus labrax) (76 +/- 16 g) were studied in an experimental tank-based flow through system. Performance, stress response and resistance to virus infection were analysed under five stabilized rearing densities: 10, 20, 40, 70 and 100 kg m(-3). Water quality parameters (CO2, total ammonia nitrogen and O-2) were measured and maintained close to the recommended values for farmed sea bass by adjusting water renewal exchange. No significant differences were observed between density treatments, neither on stress response (cortisol) nor susceptibility to nodavirus. With regards to biological performances, the daily feed intake and specific growth rate were significantly lower in fish reared at... |
